PROBLEM TO BE SOLVED: To provide such a protective group to a basic site of 2-thiouridine as to enable suppression of adverse reaction caused by an oxidizing agent in oligonucleotide synthesis. SOLUTION: The 2-thiouridine derivative is represented by formula (I) (wherein A1, A2 and A3 are the same or different and they are each hydrogen, a 1-6C alkyl, a 1-6C alkyl which may have a substituent group, a 1-7C acyl group, a silyl group which may have a substituent group, a triphenylmethyl group which may have a substituent group on an aromatic ring or a phosphoramidite group; B1 and B2 are the same or different and they are hydrogen, a 1-6C alkyl group, a 1-6C alkoxy group, an alkylamino group having one or two of hydrogen atoms or 1-6C same or different alkyl groups on the nitrogen, a 1-3C alkyl halide group having 1-7 halogen atoms, a cyano group, a nitro group, a fluorine atom, a chlorine atom or an iodine atom; C exists at 3N position or 40 position of a pyrimidine ring and C represents a p-nitrophenylethyl group, a 2-cyanoethyl group or a trialkylsilyloxymethyl group).
