摘要 PROBLEM TO BE SOLVED: To provide a method for quantitatively analyzing the bonding strength of composite of protein and DNA, and an analyzer therefor. SOLUTION: This analyzing method includes a process 1 for preparing a DNA-protein composite (composite), a spectrum measuring process 2 by an ESI-MS method for changing cone voltage (V), a process 3 for plotting a relative intensity ratio with respect to V from a spectrum to obtain the dissociation curve of the composite in a gaseous phase, a process 4 for repeating the processes 1-3 with respect to at least one kind of composite of DNAs having different arrangements or proteins, a process 5 for calculating "V[Vg (50%)]" showing the dissociation of 50% of the composite in the gaseous phase from the dissociation curve obtained through the processes 1-4, a process (6) for forming a calibration curve showing the relation of the difference between Vg(50%), wherein the free energy change quantity (ΔG) accompanying the formation of the composite becoming a standard, and the reference value ofΔG accompanying the formation of the composite in a solution and a process 7 for determining a dissociation constant with respect to other composite from Vg(50%) using the calibration curve obtained in the process 6.
