<P>PROBLEM TO BE SOLVED: To provide an oligonucleotide adaptor by which a flanking region of a well-known DNA sequence can be isolated; and to provide a primer. <P>SOLUTION: The adaptor comprises a long-chain oligonucleotide and a short-chain oligonucleotide containing a base sequence complementary to the base sequence of the 3' terminal region of the long-chain oligonucleotide. The 5' terminus of the short-chain oligonucleotide is a protruding end comprising a partial sequence of a base sequence complementary to the base sequence of a 5' protruding end caused by the digestion by a restriction enzyme causing the 5' protruding end. Each of the oligonucleotides does not form a 3'-terminal homodimer, and does not form an intramolecular hairpin structure because when the most stable homodimer is formed, the free energy is ≥-6.5 kcal/mol (in the long-chain oligonucleotide) and ≥-4.9 kcal/mol (in the short-chain oligonucleotide).
