PROBLEM TO BE SOLVED: To provide a method for separating and purifying nucleic acid, which has excellent separation performance, improved cleaning efficiency, is simple and rapid, has excellent automation and miniaturization suitability and in which a porous membrane having substantially the same separation performance and being mass produced is used. SOLUTION: The method for separating and purifying nucleic acid comprises a process for using a nucleic acid adsorbing porous membrane being a porous membrane on which nucleic acid is adsorbed by interaction substantially not associated with an ionic bond and a sample solution containing nucleic acid which is obtained by using a process (I) for injecting a specimen containing a cell or a virus to a container, a process (II) adding a nucleic acid solubilizing reagent containing a nucleic acid splitting enzyme, a chaotropic salt and a surfactant to the container and mixing the specimen with the nucleic acid solubilizing reagent, a process (III) for incubating the obtained mixed solution and a process (IV) for adding a water-soluble organic solvent to the incubated mixed solution.
