摘要 PURPOSE:To surely and efficiently separate the C-terminal fragment of a protein in high efficiency by specifically racemizing the C-terminal amino acid of a protein, fragmentizing the protein, treating the fragment with a carboxypeptidase and selecting a fragment unchanged by the treatment. CONSTITUTION:The carboxyl-terminal fragment of a protein is surely and efficiently separated in high sensitivity by specifically racemizing the carboxyl terminal amino acid of a protein, fragmentizing the polypeptide by digesting with trypsin, etc., treating the obtained fragment with a carboxypeptidase, subjecting the fragment before and after the carboxypeptidase treatment to chromatography and selecting the fragment having unchanged eluting position to obtain the fragment free from change of molecular weight by the carboxypeptidase treatment. When there are plural fragments free from change of molecular weights, the presence of D-amino acid in each fragment is examined and the fragment having D-amino acid is selected as the carboxyl-terminal fragment.
