The present invention relates to the molecular characterization of the hipA gene and the hipA7 mutant thereof. The present invention additionally provides compositions and methods for the development of hipA and mutants thereof as negative and positive selection vectors. The present invention also relates to the cloning of nucleic acid fragments using such negative and positive selection vectors by conventional recombinant DNA and recombinational cloning methods such as those employing recombination and/or topoisomerase proteins.
