Detecting prokaryotic RNA (I) comprises providing (I); reverse transcription by introduction of an RNA polymerase recognition site (RS) to produce first-strand cDNA (II); converting (II) to double-stranded cDNA (III); in vitro transcription from (III), and detecting the (I) produced. An independent claim is also included for a reagent kit for the new method comprising reagents for: (a) synthesis of (II) from (I); (b) synthesis of (III) from (II); and (c) in vitro transcription of (III).
