Immunomodulatory polynucleotides and methods of using the same

The composition of an immunomodulatory polynucleotide is described which comprises an immunostimulatory sequence with the formula: 5Æ-X1X2AX3CGX4TCG-3Æ (SEQ ID NO:62) wherein X1 is T, G, C or Z; Z is 5-bromocytosine; X2 is T, G, A or U; X3 is T, A or C; X4 is T, G or U; and wherein the sequence is not 5Æ-TGAACGTTCG-3Æ (SEQ ID NO:63) or 5Æ-GGAACGTTCG-3Æ. The use of the composition is also described such as in the manufacture of a medicament for treating infectious diseases and increasing interferon levels.
