PROBLEM TO BE SOLVED: To provide a method of manufacturing polyrotaxane good in yield and preferable in cost without using a polypseudorotaxane in a large excess and/or without using an activation reagent in a large excess. SOLUTION: The manufacturing method of polyrotaxane includes a clathlation process for obtaining the polypseudorotaxane in which carboxylated polyethylene glycols are clathlated in a skewered state at the opening parts of a plurality of cyclodextrin molecules by mixing the carboxylated polyethylene glycols, of which the both ends of the polyethylene glycols are carboxylated, and the cyclodextrin molecules; and a blocking process for obtaining a polyrotaxane having both ends of -CO-NH- blocking groups or -CO-O- blocking groups by reacting the polypseudorotaxane with a blocking group having a -NH<SB>2</SB>group or -OH group.
