PROBLEM TO BE SOLVED: To provide a method for separating and refining a nucleic acid by using a porous film excellent in separation performance, having good washing efficiency, capable of simply and rapidly carrying out separation and refining of the nucleic acid, excellent in automatic and miniaturizing suitability, and capable of producing the film having substantially same separation performance in large amounts. SOLUTION: This method for separating and refining the nucleic acid comprises using the porous film in which the nucleic acid is absorbed by an interaction in which ion bond is not participated and a sample solution containing the nucleic acid and obtained by a method comprising (I) a step for injecting a specimen containing a cell or a virus into a container, (II) a step for adding a chaotropic salt, a surfactant, a buffer solution and a proteolytic enzyme to the above container and mixing the specimen therewith, (III) a step for incubating a mixed solution obtained by the above step at 20°C to 70°C and (IV) a step for adding a water-soluble organic solvent to the incubated mixed solution.
