PROBLEM TO BE SOLVED: To provide a method for separating and purifying a nucleic acid with a porous membrane having an excellent separation performance and a good washing efficiency, capable of being simply and rapidly be used, having excellent automating and down-sizing suitability, and capable of being produced in a large amount in a state having the substantially same separation performance. SOLUTION: This method for separating and purifying the nucleic acid, comprising (1) a process for passing a specimen solution containing the nucleic acid through the nucleic acid-adsorbing porous membrane to adsorb the nucleic acid on the porous membrane, (2) a process for passing a washing liquid through the nucleic acid-adsorbing porous membrane to wash the porous membrane in the state adsorbing the nucleic acid, and (3) a process for passing a recovering liquid through the nucleic acid-adsorbing porous membrane to detach the nucleic acid from the porous membrane, is characterized in that the nucleic acid-adsorbing porous membrane is a porous membrane which adsorbs the nucleic acid by an interaction substantially not participating in an ionic bond, and the recovery of the separated and purified nucleic acid is≥50%.
