Disclosed are a novel Geobacillus thermodenitrificans CBG-A1 strain having thermostable L-arabinose isomerase activity, the said enzyme, and a method of preparing tagatose. The strain produces a thermostable L-arabinose isomerase that is capable of stably catalyzing an enzyme reaction even at high temperature. The tagatose production method is characterized in that enzyme immobilization is achieved by attaching an arabinose isomerase onto a carrier selected from among sodium alginate, alumina and silica and crosslinking the enzyme molecules using glutaraldehyde. The strain, the thermostable arabinose isomerase produced by the strain and the tagatose production method are useful for the mass production of tagatose.
