PROBLEM TO BE SOLVED: To provide a method for the separation and purification of a nucleic acid by using a porous membrane having excellent separation performance, high cleaning efficiency, enabling easy and quick operation, suitable for automation and size reduction and producible in a mass to obtain the membranes having essentially the same separation performance. SOLUTION: The method for the separation and purification of nucleic acid contains (1) a step to pass a specimen solution containing a nucleic acid through a porous membrane capable of adsorbing nucleic acids to adsorb the nucleic acid in the porous membrane, (2) a step to pass a cleaning liquid through the nucleic acid adsorbing porous membrane to clean the membrane containing adsorbed nucleic acid and (3) a step to pass a recovery liquid through the porous membrane to desorb the nucleic acid from the porous membrane. The porous membrane adsorbs the nucleic acid by an interaction essentially free from the participation of ionic bond, and the time necessary from the injection of the specimen liquid containing the nucleic acid to the collection of the nucleic acid out of the nucleic acid separation and purification cartridge is≤10 min.
