PROBLEM TO BE SOLVED: To provide a method for preparing high-concentration prion protein that is useful for a challenge test of prion protein, homogeneous and colorless and transparent and has a sufficient infectivity titer. SOLUTION: The method for separating and cooling prion protein comprises a process for filtering a prion protein solution through a membrane filter composed of a porous hollow fiber through which a solid smaller than prion protein is passed and a process for releasing prion protein from the membrane filter by reversely sending a recovery solution not to coagulate prion protein to the membrane filter having caught a virus.
