PROBLEM TO BE SOLVED: To provide a method for separating and refining a nucleic acid by using a porous film excellent in separation performance, having good washing efficiency, capable of simply and rapidly carrying out separation and refining of the nucleic acid, excellent in automatic and miniaturizing suitability and capable of producing the film having substantially same separation performance in large amounts. SOLUTION: The method for separating and refining the nucleic acid comprises (1) a step for absorbing the nucleic acid into the porous film by passing a sample solution containing the nucleic acid through a nucleic aid-absorbing porous film, (2) a step for passing a washing liquid through the nucleic aid-absorbing porous film and washing the porous film in a state in which the nucleic acid is absorbed and (3) a step for desorbing the nucleic acid from the interior of the porous film by passing a recovering liquid through the nucleic acid-absorbing porous film. In the method for separating and refining the nucleic acid, the nucleic acid-absorbing porous film is a porous film in which the nucleic acid is absorbed by interaction in which ion bond is not substantially participated and a sample solution containing the nucleic acid is brought into contact with either one side of the nucleic acid-absorbing porous film and passed through the other side of the porous film at a flow rate of 2 to 1,500μL/sec per cm<SP>2</SP>area of the film.
