PROBLEM TO BE SOLVED: To separate and purify a nucleic acid from a specimen containing the nucleic acid in a good separation performance, simply, quickly and as capable of being automatized. SOLUTION: This method for separating and purifying the nucleic acid is provided by using a nucleic acid-separating and purifying cartridge housing a nucleic acid adsorptive porous membrane in the nucleic acid-separating and purifying cartridge having at least 2 openings, and a device for generating a pressure difference between at least 2 openings in the nucleic acid-separating and purifying cartridge to perform the adsorption and desorption of a specimen solution containing the nucleic acid including a proteinase.
