PROBLEM TO BE SOLVED: To provide a method for purifying a nucleic acid, by substituting a substance which is used in an aqueous adsorption solution for accelerating bonding of the nucleic acid to a substrate, such as an inorganic support material, with another substance. SOLUTION: This method for purifying the nucleic acid includes processes (a) to (d) and, optionally, a process (e) as follows: a process (a) for making the nucleic acid which is derived from a composition containing (i) an aqueous buffer solution, (ii) a salt of a high concentration, (iii) a water-miscible non-acidic organic compound having a functional group expressed by formula I (W is C or S; and Z is O or N), and (iv) the nucleic acid adsorbed on the substrate; (b) a process for, optionally, washing the substrate containing the adsorbed nucleic acid by a washing solution; (c) a process for making the nucleic acid desorbed from the substrate, by contacting the substrate containing the adsorbed nucleic acid with a solution containing the salt of which the concentration is lower when compared with the composition in the process (a); (d) a process for purifying the nucleic acid, by separating a solution containing the desorbed nucleic acid from the substrate; and (e) a process for further purifying the nucleic acid, by precipitating the desorbed nucleic acid from the solution of the process (d) and then isolating the precipitated nucleic acid.
