摘要 51-Inosinic acid is prepared by culturing an adenine-requiring auxotrophic mutant of Brevibacterium ammoniagenes in an aqueous nutrient medium containing adenine, with the resulting accumulation of 51-inosinic acid in the medium and cells. Any adenine-requiring auxotroph of the said micro-organism is suitable, whether requiring other growth factors (e.g. biotin, xanthine) or not. Mutants employed in the process have the following A.T.C.C. numbers: 15187, 15188, 15189 and 15190. Conventional media containing a carbon source, a nitrogen source and minerals may be employed, with adenine added per se, or as an adenine-containing source (e.g. R.N.A., fish meal, yeast extract, corn steep liquor), under aerobic conditions with suitable pH control (e.g. CaCO3, urea, NH4OH, NaOH). Inosinic acid and, if desired, hypoxanthine which occurs as a byproduct, may be recovered by conventional means, such as ion exchange procedures, and the inosinic acid may be crystalline as the sodium salt. Examples are given for the process, using the above mutant strains of Brevibacterium ammoniagenes, cultural and reinoculated in shaken broth cultures.
