A method for identifying inhibitors of myeloperoxidase (MPO) comprises reacting MPO with hydrogen peroxide and a chloride source to generate hypochlorous acid in the presence of a potential inhibitor of MPO, reacting any formed hypochlorous acid with a suitable amine to fork the corresponding chloramines. Any unreacted hydrogen peroxide is the optionally removed. Any formed chloramine is reacted with a suitable detector molecule in the presence of iodide to form oxidizes detector molecule. The amount of formed oxidized detector molecule is determined by measuring the absorbance or fluorescence at a suitable wavelength and then identifying as inhibitors of MPO those compounds which cause the amount of formed oxidized detector molecule to be decreased. The method can also be used in a diagnostic test for myeloperoxidase activity.
