A serum of increased chloresterol content suitable as a control standard is prepared by (a) adding calcium ions to a serum of non-elevated chloresterol content, (b) adding a water-soluble salt of a sulphated polygalacturonic acid methyl ester methyl glycoside, (c) collecting the resultant beta-lipoprotein precipitate and purifying it by dialysis and (d) dissolving the precipitate in a serum of non-elevated cholesterol content to form said serium of increased cholesterol content. In the Example, a plasma (ACO plasma, containing trisodium citrate, citric acid and dextrose) is first treated with calcium acetate solution, thrombin is added to clot the fibrin and then after removal of the clotted fibrin the sodium salt of sulphated polyalacturonic acid methyl ester methyl glycoside ("Mepesulphate") is added. The precipitate is centrifuged and dialised against normal saline before adding to pooled normal serum (containing 200 mg. / 100c cholesterol) to give a high cholesteral control containing 400 mg./100 1 cholesterol. Calcium chloride may be used in place of the acetate. |
