PROBLEM TO BE SOLVED: To provide a method for sensitively and easily detecting protein slightly soluble in water or protein in a slightly extracted state by a succeeding immunoreaction while maintaining high extraction efficiency from a sample. SOLUTION: Protein slightly soluble in a sample, or protein in a slightly extracted state is extracted or solubilized by a water-soluble solvent that has a relatively high concentration and contains an ionic surfactant. Then, in a high-sensitivity and simple immunoassay, protein in the extracted liquid is directly detected by using an antibody obtained to protein in which the protein is modified by an ionic surfactant.
