PROBLEM TO BE SOLVED: To provide a light source of another wavelength or a stronger output for a special excitation process in addition to a standard laser in a laser scanning microscope. SOLUTION: In an operation method of the laser scanning microscope, an arrangement in the illumination beam path of the laser scanning microscope, has a mirror SP capable of being inserted rotatably into the optical path in order to couple at least an additional wavelength, and also has a high-speed rotatable mirror SP provided in a bright optical path for coupling at least another separate wavelength, but wherein a rotation process of a rotary mirror SP1 is synchronized with illumination control by a control unit so that at least the another separate wavelength is coupled to the illumination beam path.
