<p>A stock of recombinant AAV (rAAV) free of helper virus is produced by providing at least one accessory function for recombinant adeno-associated virus (AAV) virion production in a cell, but lacking at least one adenovirus late gene region, and comprising: (i) a sequence providing adenoviral VA RNA; (ii) the coding region of adenoviral E4 ORF (open reading frame) 6; and/or (iii) an adenoviral E2 72 kD coding region.;
It eliminates the need for a helper virus and provides efficient production of AAV without formation of infectious helper virus or other contaminants. Incorporation of other accessory functions in to the vector (e.g. E1a and E1b) allows AAV production in cells not permissive for helper virus infection.</p> |
