A protein Bos taurus testicle specific hyaluronidase PH-20 is provided, which protein is useful for cancer diagnosis, joint disorder treatment, eye operation and skin softening, is applicable to the yeast expression system in order to identify its unknown properties, and is useful for its mass-production. The protein Bos taurus testicle specific hyaluronidase PH-20 has the amino acid sequence set forth in SEQ ID NO:6. A gene encoding the Bos taurus testicle specific hyaluronidase PH-20 has the nucleotide sequence set forth in SEQ ID NO:5, wherein the core sequence of the Bos taurus testicle specific hyaluronidase PH-20 is free from any stop codon and frame shifts; and the nucleotide sequence of the Bos taurus testicle specific hyaluronidase PH-20 has 79% homology to pig, and 53 to 63% homology to rabbit, guinea pig, human, monkey, fox, mouse and norway mouse.
