UTILIZATION OF HNF-1beta FOR INSPECTING AND TREATING OVARIAN CLEAR CELL ADENOMATOUS CARCINOMA AND FOR SCREENING REMEDY

PROBLEM TO BE SOLVED: To provide a method for inspecting ovarian clear cell adenomatous carcinoma with HNF (hepatocyte nuclear factor)-1βgene and a protein encoding the gene, and to provide a method for screening a remedy for the disease. SOLUTION: A gene giving an expression level difference between ovarian clear cell adenomatous carcinoma (CCC) cells and non-CCC cells was analyzed with an oligonucleotide array, and HNF-1βgene expressing and exasperating only in the CCC cells was found. It was confirmed that the HNF-1βgene expressed and exasperated in both a mRNA level and a protein level by an immunoblot analysis method and by an immune structure dyeing method. When the significance of the expression of the HNF-1βin the CCC cells was examined with RNAi, it was clarified that the decrease of the HNF-1βinduced the apoptosis-like cell death of CCC. From the above results, it is shown that the HNF-1βcan be used for diagnosing the CCC and screening remedies for treating the disease. COPYRIGHT: (C)2005,JPO&NCIPI
