A composition for prevention and treatment of hepatitis B comprising the extract of African Phellinus linteus is provided, which African Phellinus linteus extract inhibits growth of hepatitis B virus without side effects and drug resistance induction by long term dosage. The composition for prevention and treatment of hepatitis B comprises the extract of African Phellinus linteus, wherein the African Phellinus linteus is encoded by a gene having the nucleotide sequence set forth in SEQ ID NO:1; the African Phellinus linteus extract is the crude extract or nonpolar solvent extract; the crude extract of African Phellinus linteus is prepared with water or methanol; the nonpolar solvent extract is prepared with hexane or ethyl acetate; and the African Phellinus linteus extract comprises polymeric polysaccharides with molecular weight of 12,000 dalton or polymeric protein with molecular weight of 12,000 dalton.
