PROBLEM TO BE SOLVED: To provide a C5-substituted deoxycytidine derivative or an N6-substituted deoxyadenosine derivative enzymically synthesizing modified DNAs especially required for DNA analysis with a DNA chip and to provide a method for synthesizing nucleic acid using the same. SOLUTION: The C5-substitured deoxycytidine derivative is characterized in that the derivative is represented by general formula äwherein, R denotes an NHX; and X denotes a hydrogen atom, an alkynyl group, an alkenyl group, an alkynyl group or an aryl group, a COCF<SB>3</SB>group, a COCH<SB>3</SB>group, a carbonylmethylimidazole group, a CH(=NH)NH<SB>2</SB>, a biotinyl group, various amino acids bound through amide bonds or a (CH<SB>2</SB>)<SB>2</SB>N[(CH<SB>2</SB>)<SB>2</SB>NH<SB>2</SB>]<SB>2</SB>group}.
