摘要 PURPOSE: A method for mass production of Hepatitis B virus vaccine is provided, which mass production of Hepatitis B virus vaccine is accomplished by inhibiting glycosylation of prosequence and HBV pre-S antigen protein. CONSTITUTION: The method for mass production of Hepatitis B virus vaccine comprises the steps of: (1) preparing a gene encoding a recombinant HBV pre-S antigen protein in which amino acids in the N-glycosylation site of the HBV pre-S antigen protein is substituted; (2) constructing an expression vector of the recombinant HBV pre-S antigen protein containing a signal peptide, prosequence and the gene encoding the recombinant HBV pre-S antigen protein; (3) transforming a host cell with the expression vector; and (4) culturing the transformed host cells, wherein the recombinant HBV pre-S antigen protein has the amino acid sequence set forth in SEQ ID NO:2 or SEQ ID NO:3; the host cell is selected from Saccharomyces, Kluyveromyces, Schizosaccharomyces, Pichia and Hansenula; and the recombinant prosequence has the amino acid sequence of SEQ ID NO:5.
