FIELD: medicine, analytical chemistry. ^ SUBSTANCE: assay of blood trace is carried out in an aqueous-ethanol medium (5-20 vol.% of ethanol) in concentrations of 3,3',5,5'-tetramethylbenzidine (TMB), (4-8) x 10-4 M, H2O2, -.1-0.4 M in the presence of 10-4 - 10-2 M of ethylenediaminetetraacetate. In the presence of blood trace the solution acquires a blue-green color and iron ions interfering for assay are masked by ethylenediaminetetraacetate. The parent TMB solution is prepared in ethanol that provides stability of solutions for above one month and they retain sensitivity for blood assay. The solution for carrying out the reaction is prepared before analysis immediately by mixing 5-20 volume parts of TMB ethanol solution with 80-95 volume parts of H2O2 an aqueous solution and ethylenediaminetetraacetate. This provides the stability of TMB solutions. Method is simple, safety and shows the selective capacity for assay of blood trace in the presence of iron ions. ^ EFFECT: improved assay method. ^ 2 tbl, 2 dwg, 3 ex |
