A biological fluid processing device includes a multiple biological fraction container (8) enclosing a liquid biological fraction. A rigid cassette (3) contains at least one biological fluid processing chamber (22). The biological fluid processing chamber (22) includes an affinity media chamber (24), a gel media chamber (30), a concentration chamber (38) and a collection chamber (34). Tubing (31) in fluid communication with the container (8) and the biological fluid processing chamber (22) is selectively compressed to provide a valving function. A programmable instrument (2) receives the cassette (3) and regulates the valving to control the floe of the biological fluid fraction through the device.
