METHOD OF COLLECTING ERYTHROCYTES AND METHOD OF ASSAYING SORBITOL

It is intended to provide a method of conveniently assaying sorbitol in erythrocytes and an assay kit. To achieve this object, it is also intended to provide a method of collecting erythrocytes without a need for centrifugation and a method of conveniently assaying sorbitol which is appropriate for assyaing an erythrocyte sample obtained by the above-described method of collecting erythrocytes. Namely, erythrocytes are conveniently collected from whole blood by a method comprising adding the blood to an erythrocyte-holding layer capable of holding erythrocytes in the blood, allowing the serum or plasma components to penetrate into an absorption layer which is provided in contact with the erythrocyte-holding layer and capable of absorbing the serum or the plasma, and then collecting the erythrocytes from the erythrocyte-holding layer holding the erythrocytes into a solution. In the sorbitol assay, sorbitol is quantified by adding sorbitol dehydrogenase, a fluorescence substrate, NAD or NDAD and diaphorase to the erythrocytes and, after reacting, measuring the fluorescent substance thus formed.
