A method for producing a transgenic non-human mammal comprising a modified pre-selected endogenous wild-type sequence is disclosed, wherein the method comprises: a) Introducing into a zygote a composition comprising a recombinase and a first and a second single-stranded substantially complementary targeting polynucleotide each having a homology clamp substantially corresponding to or substantially complementary to a pre-selected wild-type target DNA sequence, wherein said pre-selected wild-type target DNA sequence is modified by homologous recombination with at least one of said first or second substantially complementary targeting polynucleotides, and wherein the modification is selected from the group consisting of a substitution, insertion and deletion of at least one nucleotide in said pre-selected wild-type target DNA sequence, and wherein the pre-selected wild-type target sequence encodes for an ion-channel, a G-protein coupled receptor (GPCR), an immunoglobin, a growth factor, an enzyme, or a milk protein; and b) Generating said transgenic non-human mammal from said zygote.
