<P>PROBLEM TO BE SOLVED: To provide a new endo-β-(1→6)-galactanase which is useful for elucidating the functions of AGP having various physiological functions such as the growth control function of pollen tubes and the growth and reproduction control of the plants and whose applications to transformed plants such as male sterility and to food processing, medicines and protoplast-making enzymes can be expected, and to provide a gene for the endo-β-(1→6)-galactanase. <P>SOLUTION: The β-(1→6)-galactanase is purified from a commercially available cellulase preparation, Onozuka R-10, 57 times, and then analyzed with a galactan originated from Prototheca zopfii. The enzyme exhibits the maximum activity at pH 4.3, and specifically hydrolyzes β-(1→6)-galactooligosaccharides having polymerization degrees of ≥3 and their derivatives having 4-O-methyl-glucosyl groups or glucosyl groups at the non-reducing terminals. The perfect length cDNA of the enzyme was produced by 3'- and 5'-RACE-PCR, while using a RNA originated from T. viride sporophyte. <P>COPYRIGHT: (C)2005,JPO&NCIPI |
