<p>A process is provided for mixed culturing of microbial (bacterial, yeast, or fungal) host cell strains that utilize more than one carbohydrate source as a substrate, comprising: (a) culturing in a single vessel the strains, all of which are genetically marked to lack their native ability to use a set of n-1 carbohydrate sources as substrates, n being number of strains, which set of carbohydrate sources is different for each genetic marking employed; (b) diluting an isolated sample of culture until a countable number of viable cells will be present when the sample is plated; (c) plating the diluted sample onto culture media supplemented with one carbohydrate source only utilized by one of the strains as a substrate, and with no other carbohydrate sources; (d) incubating the plated cells at a temperature and for a time sufficient for any positive colony to grow to a detectable level; and (e) comparing growth or cell population changes of the different strains by enumerating the colonies growing on the supplemented culture media and comparing the number of colonies on each of the carbohydrate sources.</p> |
