PROBLEM TO BE SOLVED: To simply detect whether a gram-negative or gram-positive bacterium exists or not in a sample in good operability from enzymatic activity or metabolism or the change of living bacterial numbers. SOLUTION: When carrying out detection of the gram-positive bacterium, benzalkonium chloride in a prescribed amount is added to the bacterium and a detection reagent is added thereto and the amount of a produced labeled substance is determined. When carrying out detection of the gram-negative bacterium, a detection reagent is added to the bacterium without adding the benzalkonium chloride and the amount of a labeled substance to whole bacterium cell is measured and a difference from the measuring result in the case of the gram-positive bacterium is calculated and whether the gram-negative bacterium exists or not is determined from the difference.
