PROBLEM TO BE SOLVED: To extremely improve the stability, activity and convenience of using operation of a reagent used for isothermal amplification of a nucleic acid by solving the problems in which the amplification reaction proceeds under operation of the reagent and under preservation. SOLUTION: This method involves carrying out the gene amplification in the coexistence of at least one of the following oligonucleotides having sequences competing with a target gene sequence against a chimera oligonucleotide primer used in the amplification: The oligonucleotide has the ratio of the Tm value between the chimera oligonucleotide primer and a nucleic acid sequence of the target gene substantially complementary to the base sequence of the primer, to the Tm value between the oligonucleotide and the chimera oligonucleotide primer, regulated within the range of 1.2-3.3, and also has its 3' terminus blocked so as not to be extended.
