PROBLEM TO BE SOLVED: To provide a measurement method for signal-transmitting activity through G-protein conjugation receptor (GPCR) using animal cells, such as oocytes of Xenopus laevis.
SOLUTION: The oocytes that are extracted from Xenopus laevis and selected by a usual method are co-introduced with an RNA encoding GPCR and an RNA encoding a chimera G protein, for example, G11 or Gq and G14, G15 or G16. After introduction of the RNA, the oocytes are cultured for a normal period and ligand candidate substances are added to the culture mixture and the activity is measured. According to this method, whether the candidate is a ligand or not can be judged by only one measurement. Thus, the measurement method for identification of the GPCR ligand having unknown functions is simplified and accelerated.
COPYRIGHT: (C)2005,JPO&NCIPI |
