A method for generating and capturing peptides on a target surface. The method comprises electrophoresing a protein on a high resolution reversibly polymerized polyacrylamide gel and vacuum depositing the electrophoresed protein on a digestion membrane for digestion into peptides. The digestion membrane is a substantially chemically inert membrane having a proteolytic enzyme immobilized thereon. The method further comprises vacuum depositing the generated peptides on the target surface. A target surface for use in a mass spectrometer comprises a porous plate that does not ionize or volatize in the charge conditions of the mass spectrometer. In its preferred form, the porous plate has a coating of organic matrix and nitrocellulose. Further, the invention relates to a high vacuum stage for procuring biopolymers attached to a membrane and depositing them on a porous plate target surface. |
