PROBLEM TO BE SOLVED: To provide a flow fluorescence apparatus, and to provide a method of using two-photon excitation and/or a confocal optical setup. SOLUTION: The optical setup is optimum for counting small fluorescent biological particles. An active confocal volume is diffraction-limited, which is considerably smaller than the volume of a flow channel. It has been found that the concepts of excitation and detection are very effective for eliminating a background signal. An objective lens, having a high numerical aperture for focusing a laser and collecting fluorescence, is used, which limits an active volume for measurement to a diffraction-limited volume nearly equal to a volume on the order of femtoliter.
