PROBLEM TO BE SOLVED: To provide an efficient and economical method for producing bivalent antibodies especially F(ab')2molecules, and to provide a method for enabling to modify bivalent and bi-specific F(ab')2molecules. SOLUTION: The method for producing an Fv polypeptide containing immunoglobulin H chain Fv region and immunoglobulin L chain Fv region, wherein the H chain or the L chain contains an unpaired cysteine residue as a free thiol, comprises manifesting and secreting the Fv polypeptide in periplasm of a recombination colon bacillus cell culture solution and collecting the polypeptide under a condition of practically maintaining the cysteine residue as the isolated thiol.
