A cellophane agar medium is made of a cellophane piece with groove portions on at least one surface thereof and a nutritious agar formed on the surface of the cellophane piece via the groove portions. Then, the cellophan e agar medium is disposed under wet condition, and a microbe is inoculated and cultivated on the cellophane agar medium. A fixing solution is added to the cellophane agar medium to fix the microbe to the cellophane agar medium. Thereafter, a given sample is made of the cellophane agar medium with t he microbe and then, observed with a scanning electron microscope or a transmission electron microscope.
