PROBLEM TO BE SOLVED: To provide a method for purification of chondroitin lyase enzyme, a DNA sequence encoding the chondroitin lyase enzyme and to provide a purified chondroitin lyase enzyme useful as a pharmacological reagent. SOLUTION: The invention relates to the method for production of two highly purified enzymes degrading chondroitin sulfate polysaccharide, comprising a multi-step purification method incorporating cell disruption, cation exchange chromatography, affinity chromatography, hydroxyapatite chromatography, high resolution ion exchange chromatography and size exclusion, (wherein the enzyme is a protein with 77,000±5,000 Dalton of molecular weight, degrading chondroitin sulfates A and C, and a protein with 55,000±2,300 Dalton of molecular weight and degrading dermatan sulfate).
