PROBLEM TO BE SOLVED: To provide a genetic mutant animal widely useful as a model of morbid states such as senescence acceleration and immunological deficiency.
SOLUTION: A nonhuman mammal, especially a mouse is deficient in a gene encoding an APEX2 enzyme that is an AP endonuclease. The mammal is prepared by using an ES cell (an embryo-stem cell) in which the gene encoding the APEX2 enzyme is replaced with a function-deficient type gene. The mammal assumes the growth retardation caused by natural DNA injury, has high sensitivity to lesions caused by the injury and remarkably reduces cells in charge of immunity Thereby, the mammal is useful as the model of the plurality of morbid states associated with the senescence acceleration, immunological deficiency, etc.
COPYRIGHT: (C)2005,JPO&NCIPI |
