An analog of human amyloid plaque core protein protease inhibitor wherein the amino acid corresponding to arginine at position 13 in the sequence <MATH> is substituted with an aromatic amino acid, said analog exhibiting chymotrypsin inhibitory activity or is substituted with a neutral hydrophobic amino acid, said analog exhibiting human elastase inhibitory activity and method of producing them. Use of the protease inhibitor in the manufacture of a medicament for use in treating acute pancreatitis and method of recovering purified protease inhibitor.
