METHOD FOR PRODUCING NUCLEOSIDE

PROBLEM TO BE SOLVED: To provide a method for producing a nucleoside having high general-purpose properties using a 1-phosphated saccharide which is an anomeric mixture readily synthesizable at a low cost as a reaction substrate. SOLUTION: An exchange reaction of a phosphate group of the 1-phosphated saccharide with a base is anomer-selectively carried out when a reaction of the anomeric mixture of a nonnatural type 1-phosphated saccharide with the base is conducted by actions of a phosphorylase activity. Furthermore, metal cations forming a sparingly water-soluble salt with phosphate ions are made to exist in a reaction liquid to thereby precipitate the phosphate ions which are by-products of the reaction as the sparingly water-soluble salt. As a result, yield of reaction is improved by shifting the equilibrium of the reaction in the direction of nucleoside synthesis. A mixture containing a large amount of one of optical isomers of the nucleoside can be obtained by utilizing a difference in rate of the exchange reaction. COPYRIGHT: (C)2005,JPO&NCIPI
