PROBLEM TO BE SOLVED: To provide a method for measuring calcium by making use of the activation of phospholipase D by calcium ion. SOLUTION: A reagent for measuring calcium in specimens is provided, containing the phospholipase D≥50% in the ratio of a phospholipase D isozyme with a molecular weight of about 57 kilodalton and a compound of formula(I)[wherein, X1 is a chromogenic group; and X2 is H, a chromogenic group or a (substituted) alkyl or phenyl] as a substrate for the phospholipase D.
