It is intended to provide a method of designing normally orthogonalized sequences, a method of producing nucleic acids which are normally orthogonalized sequences and nucleic acids obtained thereby. Namely, a method of designing normally orthogonalized sequences which comprises: first or preliminarily constructing a plural number of base sequences at random; determining the mean of the melting points thereof; obtaining candidate sequences based on a threshold restricted to a temperature range corresponding to the mean ± 1sigma; and obtaining normally orthogonalized sequences from the candidate sequences depending on, as an indication, whether or not being a sequence reacting independently.
