<P>PROBLEM TO BE SOLVED: To provide a method for removing efficiently serum albumin and immunoglobulin existing in large quantities in serum, and a buffer capable of solubilizing a serum protein component. <P>SOLUTION: In this pretreatment method of the serum, a serum sample to be subjected to the pretreatment is subjected to ammonium sulfate fractionation by 55-65% saturated ammonium sulfate, and an acquired precipitate fraction is refined by using a carrier formed by bonding an affinity adsorbent relative to the serum albumin to a support and a carrier formed by bonding an affinity adsorbent relative to the immunoglobulin to a support. Preferably, the affinity adsorbent relative to the serum albumin is Chibacron Blue F3GA, and the affinity adsorbent relative to the immunoglobulin is protein A, and the composition of lysisbuffer is 5-8M urea, 1.5-2.5M thiourea, 2-4 wt.% 3-[(3-colamid propyl)-dimethylanmmonio]-1-propane sulfonic acid, 0.5-2 wt.% Toriton X-100, 0.2-0.6 wt.% amphoteric carrier, and 20-70nM dithiothreitol. <P>COPYRIGHT: (C)2005,JPO&NCIPI |
