HUMAN THROMBOPOIETIN DERIVATIVES HAVING IMPROVED BIOLOGICAL ACTIVITY AND INCREASED HALF LIFE AND METHOD FOR PREPARING SAME

PURPOSE: Human thrombopoietin derivatives and a method for preparing the same derivatives are provided. The derivatives have improved biological activity and increased half life, so that they can be useful for treatment of platelet-reducing disease by anticancer treatment or bone marrow transplantation. CONSTITUTION: The human thrombopoietin derivatives in which 82th amino acid of glycine, 83th amino acid of proline and 117th amino acid of arginine residue in the amino acid sequence of natural human thrombopoietin of SEQ ID NO:2 are substituted with asparagine, alanine and asparagine, respectively, thereby N-sugar chain is introduced into the human thrombopoietin derivatives are provided. A gene encoding the recombinant human thrombopoietin derivatives has the nucleotide sequence of SEQ ID NO:17. An expression vector pCMV-dhfr-gTP01/S1 comprises the human thrombopoietin derivative gene of SEQ ID NO:17. A transformed mammal cell line(KCTC 10459BP) is produced by transforming with expression vector pCMV-dhfr-gTP01/S1. The method for preparing the human thrombopoietin derivatives comprises culturing the transformed mammal cell line(KCTC 10459BP).
