发明名称 METHOD FOR ASSAYING VANISHMENT OF DOUBLE-STRANDED STRUCTURE OF NUCLEIC ACID AND KIT THEREFOR
摘要 PROBLEM TO BE SOLVED: To provide a new method for assaying the vanishment or unwinding activity of the double-stranded helical structure of a nucleic acid or the ability of a single-stranded nucleic acid-specific binding protein, and to provide a simple kit for the method with high quantitativeness and reproducibility. SOLUTION: The new method comprises the steps of (1) preparing a 1st single-stranded nucleic acid probe having a fluorescence energy donor on 5' or 3' end and a 2nd single-stranded nucleic acid probe complementary to the 1st single-stranded nucleic acid probe and having a fluorescence energy acceptor on the opposite end, (2) causing fluorescence resonance energy transfer(FRET) with the pair of nucleic acid probes as a double-stranded nucleic acid, and (3) measuring a fluorescence spectrum change by the cancellation of the FRET associated with the vanishment of the double-stranded structure of the double-stranded nucleic acid.
